It demonstrates the presence of metachromatic granules found in Corynebacterium diphtheriae.The granules stain purple -black against light green counterstained cytoplasm. It helps to distinguish Corynebacterium diphtheriae from most of the short nonpathogenic diphtheroids which lack granules.
